<p style="text-align: center;">SONG Chunhua, DING Xu, WANG Baoye, FU Jiaming, XU Hongfei, GONG Weizhi, DONG Qinjian,&nbsp; LI Xiaoyan, GUO Dan, JIN&nbsp; Yan-Hong,&nbsp; YAN Jun,KANG Lianru</p><p style="text-align: center;">（1 Heilongjiang University Of Chinese Medicine, Heilongjiang Harbin 150040；2 Yilongxian people&#39;s hospital,&nbsp; &nbsp;Yilong 637600）</p><p style="text-align: justify;"><strong>Abstract</strong>:&nbsp; <strong>Objective</strong>:&nbsp; To&nbsp; investigate&nbsp; the&nbsp; molecular&nbsp; mechanism&nbsp; by&nbsp; study&nbsp; of&nbsp; Xinshu&nbsp; electrical acupuncture preconditioning&nbsp; on&nbsp; the&nbsp; open&nbsp; regulation&nbsp; mechanism&nbsp; of&nbsp; mitochondrial&nbsp; permeability transition pore in rat ischemic myocardium&nbsp; .&nbsp; Methods:&nbsp; Rats divided into blank group, model group, xinshu acupuncture preconditioning group , ischemic preconditioning group by random number table&nbsp; corresponding.&nbsp; Duplicate&nbsp; myocardial&nbsp; ischemia&nbsp; and&nbsp; reperfusion&nbsp; injury&nbsp; model&nbsp; by&nbsp; alive&nbsp; rat heart&nbsp; ligation&nbsp; of&nbsp; left&nbsp; anterior&nbsp; descending&nbsp; coronary&nbsp; artery .&nbsp; Blank&nbsp; group:&nbsp; Rats&nbsp; are&nbsp; fixed&nbsp; with acupuncture preconditioning group for 30 minutes but not acupuncture before molding;once per day,continue 15 days. Model group:&nbsp; Rats are fixed with blank group before molding; No other factors&nbsp; were&nbsp; imposed&nbsp; on&nbsp; the&nbsp; whole&nbsp; experiment&nbsp; except&nbsp; myocardial&nbsp; ischemia reperfusion.Acupuncture preconditioning group: Acupuncture 15 days until molding; once per day, 30 min every time,then MIRI molding.Ischemic preconditioning group: Dealing with rats the same as&nbsp; the&nbsp; blank&nbsp; group&nbsp; before&nbsp; molding; To block&nbsp; blood&nbsp; flow 5&nbsp; min&nbsp; then&nbsp; perfusion&nbsp; 5&nbsp; min&nbsp; &nbsp;before coronary&nbsp; artery&nbsp; ligation&nbsp; on&nbsp; molding.&nbsp; Repeated&nbsp; 3&nbsp; times&nbsp; in&nbsp; a&nbsp; row,&nbsp; a&nbsp; total&nbsp; of&nbsp; 30&nbsp; min&nbsp; ischemic preconditioning.&nbsp; Detect&nbsp; myocardial&nbsp; cell&nbsp; mitochondrial&nbsp; membrane&nbsp; permeability&nbsp; transition&nbsp; pore opening degree after myocardial ischemia molding success 0 h, 24 h, 48 h. <strong>Results</strong>: 1. The model group&nbsp; dyed&nbsp; myocardial&nbsp; mitochondria&nbsp; green&nbsp; fluorescent&nbsp; compared&nbsp; with&nbsp; the&nbsp; blank&nbsp; group&nbsp; was reduced obviously by laser confocal microscope observing, the experiment result shows that the mitochondrial membrane permeability transition pore activity of model group was significantly reduced&nbsp; (P &lt; 0.01). 2. Ischemic&nbsp; preconditioning&nbsp; 0h&nbsp; group&nbsp; and&nbsp; acupuncture&nbsp; preconditioning&nbsp; 0h group&nbsp; mitochondrial&nbsp; fluorescence&nbsp; intensity&nbsp; is&nbsp; significantly&nbsp; reduced&nbsp; compared&nbsp; with&nbsp; the&nbsp; model group by laser confocal microscope observing, but the indicators have no significant difference between&nbsp; ischemic&nbsp; preconditioning&nbsp; 0h&nbsp; group&nbsp; and&nbsp; acupuncture&nbsp; preconditioning&nbsp; 0h&nbsp; group&nbsp; (P&gt; 0.05).3. Acupuncture preconditioning 24h, 48h group mitochondria fluorescence intensity have a certain enhancement (P &lt; 0.05 or P&lt;0.01) observed by laser confocal microscope compared with ischemic&nbsp; preconditioning&nbsp; group&nbsp; and&nbsp; acupuncture&nbsp; preconditioning&nbsp; 0h&nbsp; group.4.&nbsp; Experiment&nbsp; data shows&nbsp; that&nbsp; acupuncture&nbsp; preconditioning&nbsp; group&nbsp; and&nbsp; ischemic&nbsp; preconditioning&nbsp; group&nbsp; both&nbsp; can reduce myocardial ischemia reperfusion injury; And perhaps acupuncture preconditioning 24&nbsp; h group is the best treatment plan.&nbsp; <strong>Conclusion:</strong>&nbsp; 1. The protection of electrical acupuncture Xinshu preconditioning for myocardial ischemia after 0h,24h,48h was different, the therapy effect enhance - peak - attenuated trend increased with time,and perhaps acupuncture preconditioning 24 h group is&nbsp; the&nbsp; best&nbsp; treatment&nbsp; plan.2.Electrical&nbsp; acupuncture&nbsp; Xinshu&nbsp; has&nbsp; protective&nbsp; effect&nbsp; for&nbsp; MIRI&nbsp; ,the mechanism may be by inhibiting mPTP opening in myocardial ischemia reperfusion injury and reducing the mitochondrial apoptosis then reduce the myocardial ischemia injury.</p><p><strong>Key words:</strong> Acupuncture and massage, Acupuncture preconditioning,&nbsp; Ischemic preconditioning,&nbsp;The laser confocal microscope, Mitochondrial permeability transition pore</p><p><br/></p>
