Effects of Electroacupuncture Pretreatment on mRNA Expression of TLR4、MyD88 and NF-кB in the PC6 Area of Rats with Myocardial Ischemia-reperfusion Injury

<p style="text-align: center;">ZHU Yi, LI Yi,ZHANG Nan, DONG Guirong</p><p style="text-align: center;">(Department of Acupuncture, Yueyang Hospital of Integrated Traditional Chinese Western Medicine, ShanghaiUniversity of Traditional Chinese Medicine,Shanghai, 200437,China)</p><p style="text-align: justify;"><strong>Abstract</strong>:&nbsp;<strong>Objective</strong>:To observe the effect of electroacupuncture(EA) pretreatment on mRNA expression of TLR4、MyD88 and NF-кB of “Neiguan” Point(PC 6)in the PC 6 area of rats with myocardial&nbsp; ischemia-reperfusion&nbsp; injury.&nbsp; <strong>Methods </strong>:&nbsp; &nbsp;The&nbsp; healthy&nbsp; wistar&nbsp; rats&nbsp; were&nbsp; randomly divided into 6 groups, i.e., the control group, the normal EA group, the sham operation group ,the model group, the EA group, the sham EA group, 8 in each group. The rats in the EA group, the normal EA group and the sham EA group were treated with&nbsp; EA (2~20 Hz in frequency, tolerated intensity of rats) at “Neiguan” (PC 6), 30 min per treatment, once a day for consecutive 5 days. The rats in the sham operation group and model group were treated with immobilization for the same&nbsp; time,&nbsp; and&nbsp; no&nbsp; EA&nbsp; was&nbsp; given. Animals&nbsp; were&nbsp; subjected&nbsp; to&nbsp; 30&nbsp; min&nbsp; of&nbsp; ischemia&nbsp; induced&nbsp; by occluding the left anterior descending artery followed by 120 min of&nbsp; reperfusion after the end of EA. The mRNA expression of TLR4, MyD88 and NF-κB in the&nbsp; PC6 area were determined with Real-time&nbsp; PCR.&nbsp; <strong>Results</strong> :&nbsp; Compared&nbsp; with&nbsp; the&nbsp; normal&nbsp; group,&nbsp; the&nbsp; ECG&nbsp; J-point&nbsp; height&nbsp; were significantly increased in the model group (P&lt;0.01), compared with the model group and&nbsp; the sham EA group ,EA preconditioning can&nbsp; significantly&nbsp; reduced ECG&nbsp; -&nbsp; J point height(P&lt;0.01); The&nbsp; mRNA expression of TLR4, MyD88 and NF-κB were significantly increased in the model group and the sham&nbsp; EA&nbsp; group&nbsp; compared&nbsp; with&nbsp; the&nbsp; control&nbsp; group&nbsp; (P&lt;0.01). EA&nbsp; preconditioning&nbsp; significantly reduced the the mRNA expression of TLR4, MyD88 and NF-κB compared with the model group and the sham EA group (P&lt;0.05).&nbsp; Conclusion:&nbsp; EA preconditioning significantly reduced the the mRNA expression of TLR4, MyD88 and NF-κB in the PC6 area of rats with myocardial ischemiareperfusion injury.</p><p style="text-align: justify;"><strong>Key words</strong>: Acupuncture, The PC6 area, Ischemia-reperfusion injury, TLR4、MyD88、NF-кB</p><p style="text-align: justify;"><br/></p>
