The Effect of Moxibustion on Proliferation and Differentiation of Neural Stem Cellsin Vascular Dementia Rat Hippocampus

<p style="text-align: center;">CUI Jingcheng, YANG Jun</p><p style="text-align: center;">(1 Graduate school, Anhui University of Chinese Medicine, Anhui Hefei 230038;2 The First Affiliated Hospital of Anhui University of Chinese Medicine, Anhui Hefei 230038)</p><p style="text-align: justify;"><strong>Abstract</strong>: <strong>Objective</strong>: To observe the effect of moxibustion at GV14, GV20 and GV24 on NSCs’ proliferation&nbsp; and&nbsp; differentiation&nbsp; in&nbsp; VD&nbsp; rats’&nbsp; hippocampus.&nbsp; Methods:Establishing&nbsp; vascular dementia&nbsp; model&nbsp; in&nbsp; rats&nbsp; by&nbsp; ligating&nbsp; right&nbsp; and&nbsp; left&nbsp; common&nbsp; carotid&nbsp; artery&nbsp; successively&nbsp; after repeatedly ischemia-reperfusion.Cultivating and passaging neural stem cells and endothelial cells respectively,&nbsp; marking&nbsp; neural&nbsp; stem&nbsp; cells&nbsp; with&nbsp; lentivirus&nbsp; mediated&nbsp; EGFP,&nbsp; and&nbsp; constructing&nbsp; cotransplanted implant in virto for celluar transplantation. 24 model rats were divided into 1 groups randomly as NE group, NEM group. And each group&nbsp; was divided into 2 parts by the treatment intervention&nbsp; period&nbsp; as&nbsp; T1&nbsp; and&nbsp; T2.&nbsp; Rat&nbsp; brains&nbsp; were&nbsp; taken&nbsp; on&nbsp; time&nbsp; after&nbsp; perfusion&nbsp; of&nbsp; 4% paraformaldehyde.&nbsp; Marker&nbsp; protein&nbsp; expression&nbsp; was&nbsp; detected&nbsp; by&nbsp; laser&nbsp; confocal&nbsp; microscope&nbsp; to analyse the moxibustion effect of neurogenesis.&nbsp; <strong>Results</strong>:The MAP expression in NE group was significantly decreased in T2 than in T1 (P<0.05). The T2 MAP expression in NEM group was significantly&nbsp; higher&nbsp; than&nbsp; in&nbsp; NE&nbsp; group&nbsp; (P < 0.05).&nbsp; The&nbsp; NeuN&nbsp; expression&nbsp; in&nbsp; NE&nbsp; group&nbsp; was significantly decreased in T2 than in T1 (P<0.05). The T2 NeuN expression in NEM group was significantly higher than in NE group (P<0.05). <strong>Conclusion</strong>: Moxibustion can promote NSCs’ proliferation and differentiation in VD rats’ hippocampus.</p><p style="text-align: justify;"><strong>Key&nbsp; words</strong>:&nbsp; Acupunctureand&nbsp; moxibustion,&nbsp; vascular&nbsp; dementia,&nbsp; neural&nbsp; stem&nbsp; cell,&nbsp; proliferation, differentiation</p><p><br/></p>
